This project originates from the hypothesis that the biophysics of lipid digestion, using different physical and chemical states of lipids, may alter lipid-lipase interactions, thusaltering the rate of FFA release in a similar manner to which Xenical, the only FDA approved drug for weight loss, functions. Xenical binds to pancreatic lipase, inhibiting its activity and reducing the amount of free fatty acids (FFA) and monoglycerides available to cross the epithelia cells entering the portal vein or lymphatic system. Once FFA have been generated by the lipolytic action of lipase, the trafficking and biological impact of FFA are very well understood. The biological impact of high-fat diets are major risk factors for obesity, Type II diabetes, hypertension, and atherosclerotic vascular disease. Although the role of elevated blood lipids has been well established as a detrimental influence on human health, there is very little understanding on how food formulation and processing affect the rate of lipid hydrolysis in the lumen. This project will evaluate whether food form, such as emulsion structure, polymorphism and fractal dimensions in colloidal fat crystal networks, fatty acid composition of lipid species and processing (i.e., cooking, drying and particle size reduction), alters lipid-lipase interactions in the lumen. These study have not occurred because they are nearly impossible to do in animal studies without extremely invasive methods. However, a sophisticated method for simulating digestion, the TIM-1, will be used to observe the influence of food form and processing on lipid metabolism. The TIM-1, available at the New Jersey Institute for Food, Nutrition and Health,is the only such unit in an academic institute in the entire country. Coupling the simulated digestion using the TIM-1 with advanced analytical tools to evaluate food form, this study will be a first-of-its-kind dealing with issues of tremendous social importance.
