Major glycolipid of human gastric content

Abstract

Although much has been learned about the glycolipids of the gastric mucosa, there is very little information available on glycolipids of the gastric content. To fill in this gap the authors have extracted glycolipids from a neutralized pool of human gastric juices collected after pentagastrin stimulation and examined them by silicic acid column chromatography and thin layer chromatography (TLC) in several different solvent systems. The acetone eluate from silicic acid column was found to contain one major and several minor glycolipid components. The yield of the major glycolipid was 0.6mg/ml native gastric juice and 0.42mg/mg protein was determined by Lowry method. Gas-chromatography and other analyses of the major glycolipid component indicated the presence of fatty acids, glycerol ethers, glucose and sulfate, and absence of sphingosine. The migration of this compound on TLC was retarded by mild alkaline methanolysis and enhanced by desulfation. Preliminary analytical results indicate that this compound is a monoalkyl-monoacyl-glyceryl-glucoside sulfate. This compound has not been as yet detected in the gastric mucosa of man.
